Protein and Polypeptide Structure There are four levels of structure found in polypeptides and proteins. The primary structure of a polypeptide protein determines its secondary, tertiary, and quaternary structures. Primary Structure The primary structure of polypeptides and proteins is the sequence of amino acids in the polypeptide chain with reference to the locations of any disulfide bonds. The primary structure may be thought of as a complete description of all of the covalent bonding in a polypeptide chain or protein. The most common way to denote a primary structure is to write the amino acid sequence using the standard three-letter abbreviations for the amino acids. For exampleà gly-gly-ser-alaà is the primary structure for a polypeptide composed of glycine, glycine, serine, and alanine, in that order, from the N-terminal amino acid (glycine) to the C-terminal amino acid (alanine). Secondary Structure Secondary structure is the ordered arrangement or conformation of amino acids in localized regions of a polypeptide or protein molecule. Hydrogen bonding plays an important role in stabilizing these folding patterns. The two main secondary structures are the alpha helix and the anti-parallel beta-pleated sheet. There are other periodic conformationsà but the à ±-helix and à ²-pleated sheet are the most stable. A single polypeptide or protein may contain multiple secondary structures. An à ±-helix is a right-handed or clockwise spiral in which each peptide bond is in the trans conformation and is planar. The amine group of each peptide bond runs generally upward and parallel to the axis of the helix; the carbonyl group points generally downward. The à ²-pleated sheet consists of extended polypeptide chains with neighboring chains extending anti-parallel to each other. As with the à ±-helix, each peptide bond is trans and planar. The amine and carbonyl groups of peptide bonds point toward each other and in the same plane, so hydrogen bonding can occur between adjacent polypeptide chains. The helix is stabilized by hydrogen bonding between amine and carbonyl groups of the same polypeptide chain. The pleated sheet is stabilized by hydrogen bonds between the amine groups of one chain and the carbonyl groups of an adjacent chain. Tertiary Structure The tertiary structure of a polypeptide or protein is the three-dimensional arrangement of the atoms within a single polypeptide chain. For a polypeptide consisting of a single conformational folding pattern (e.g., an alpha helix only), the secondary and tertiary structure may be one and the same. Also, for a protein composed of a single polypeptide molecule, tertiary structure is the highest level of structure that is attained. Tertiary structure is largely maintained by disulfide bonds. Disulfide bonds are formed between the side chains of cysteine by oxidation of two thiol groups (SH) to form a disulfide bond (S-S), also sometimes called a disulfide bridge. Quaternary Structure Quaternary structure is used to describe proteins composed of multiple subunits (multiple polypeptide molecules, each called a monomer). Most proteins with a molecular weight greater than 50,000 consists of two or more noncovalently-linked monomers. The arrangement of the monomers in the three-dimensional protein is the quaternary structure. The most common example used to illustrate quaternary structure is the hemoglobin protein. If all of the monomers in a protein are identical, there is homoquaternary structure. Hydrophobic interaction is the main stabilizing force for subunits in quaternary structure. This sounds like a lovely idea, and a very creative way to turn what is essentially a controversial garbage dump into something more positiveâ⬠¦something the public can enjoy and be proud of. The Cityââ¬â¢s choice not to relocate the landfill and go with a more environmentally sound decision was a wise one, helped enormously by public input. The Implication: Social Risk vs Economics The City of Guelph had for some time known it would have to deal with the issue of a needed landfill. In fact from 1987 discussions had been sporadically on-going with little progress, the reality being that no one wants a landfill in their backyard for all of the obvious reasons. In January of 1993 a proposed dump site north of Guelp was eliminated from further consideration after opponents managed to convince city council to throw out almost three years of work that had cost nearly $3 million. It was the beginning of a back and forth battle that would discourage county involvement and place the onus of burden on the city of Guelph. The search, beginning in 1994, would be controversial, intense and difficult. Based on records the intension of including public opinion and that and of others was, at least at first, well meaning. Ali (1999) in his study of the project writes, ââ¬Å"In order to avoid the pitting of the public against the technical consultants that had occurred in a previously unsuccessful search, Guelph City Council wished to adopt a more inclusive process in which all members of the community could voluntarily participateâ⬠(p. 1). To this end the Landfill Search Group (LSG), a Community Advisory Subcommittee of varied professionals and the (CASC), or Neighbourhood Liaison Groups made up of volunteers from all walks of life from the community and ostensibly representing the public. It would seem that things would go smoothly, but that was not the case and the public, suspicious of how t he groups were selected and would perform, formed their own counter groups who, at the outset, were set against the landfill. After the LSG announcement of five potential sites, other neighbourhood groups were formed against the landfill for environmental reasons and surprisingly, on the basis that the sites which were being considered were all in lower economic areas. In essence, the latter complaints were ââ¬Å"primarily on the basis of social equityâ⬠(Ali, 1999, p. 1) and the fact that the area was already home of a good share of the industry of the city. All of the groups came up with a list of 48 criteria on which the selection would be based, prioritized by the public which, in the end, promoted a site considerate of public health and the environment. From the beginning much of the controversy centered upon the components of priority ranking of the various aspects of the decision making process concern acceptable tradeoffs ââ¬Å"between economic and environmental impac tsâ⬠¦to both the community and to the Cityâ⬠(Ali, 1999, p. 1). Changing places
Change of places can be for a good or bad reason, but in my case was for the best for our family. Most couples, when thinking about divorcing are worried about their children, because everyone knows that divorce have some effects in children's. In my opinion divorce is about loss and change. When my parents divorced it was hard for my younger brother and l. But was a change for the best of everyone. We moved to Hidalgo. Around this change of place, as result of my parents' divorce, everything has changed, a new place to live, a new school and we had to grow up emotionally faster.My life as many peoples had good and difficult times, but I have learned about life struggles. I was sixteen years old when my parents divorced. One night, my brother was in his room, and I was in the kitchen. Our parents told us to come and sit down in the living room. They had to told us something. We all were sit in the room, my Mom said â⬠your Dad and I ââ¬Å", my Dad interrupted her, â⬠are g etting divorceâ⬠. We were in shock. The next day my Mom told me that we had to move. She decided to come here, because we can get a better education here.We came like three times in a onto for shopping, and, to visit my aunts. But it was not the same. We had lived all of our lives in Mexico, so it was a huge change for us. At the time, when we came from Mexico, we were a little family: my Mom, my little Brother and l. It was hard because we did not have a stable place to live, we stayed with our aunt while my Mom found a place to live. Every day my Mom was out looking for an apartment or house to rent. This experience was completely new for us, we had our own house In Mexico, but my parents decided to sale the house, when they divorced.After a peoples of months; finally, my Mom found an apartment of two rooms. The first couple of months were the most difficult for us. We got in a new school, without knowing English. My brother and l, took SSL classes to help us to master the la nguage. Every day when I came back from school to our new apartment which had no furniture. At that time It was only my Mom working to pay the bills without any help from my Dad, so we did not had too much In our little apartment , only the basics. As many teenagers we wanted everything. My brother was fourteen years old, so he cannot work at the time.I was sixteen years, so I decided to look for a Job to be able to buy my personal stuffs; I got a Job in an import and export agency as secretary. After six months, my little family was more stable in all terms, but my Mom got some eye problems, so she was obligated stop working. As the oldest in my little family I had to change my part time Job for a full time Job to support them, at this point, when I changed the house obligations with my Mom, I stopped to see my family that little. My Mom said ââ¬Å"sorry for give you this responsibility at this ageâ⬠I felt myself onto a tornado, this experience was totally new.I learned to n ever give up at any change. I had to grow up more quickly, be mature to distinguish the good from the bad. Sometimes, I felt sad, because I want to party with my friends from school, but I had to work a full time Job on weekends, pay bills, drive my Mom to her appointments not give up in his studies. After all, in my Junior year I noticed that if I had finish with my credits I will have the chance to leave early instead of pm in my senior year, it was like a little motivation for me to get better in school and never give up, as my mom said â⬠hard work always pays off'.Finally, I went to my senior year, I did all my credits, so I change my schedule to have more time. I get experience in import/export and brokers in my first Job, so I applied in a new agency as broker consultant, I get the Job and a raise too. Through, this years was a lot of stress for me have two Jobs, school and the responsibility of support my little family, at the time I was too busy thinking about my new r esponsibilities that I did not remember about my parents' divorce and the change of place. I accepted the changes.And now I had done with my high school, and still in college with new opportunities to face. Around this change I had to work for my family and my goals, everything was new, no body say, it was easy changing places, but my family did it. We changed of place for a better life, not for the reason we wanted but no matter if are good or bad the reason, the importance is to pick the good from the bad. Instead, I am an independent women, and I am making my Mom proud of me. From this change I realized that no matter how bad something seems you will always like the results at the end.
